Test Consists

Mycobacterial blood culture after lysis/centrifugation.

Specimen Requirements

8 - 10 mL blood submitted in adult Isolator tube; pediatric Isolator tube up to 1.5 mL blood.

Volume and Container

Isolator tube

Storage and Transport

Transport at ambient temperature to Microbiology Laboratory within one hour.

Critical Values

 Phone report of acid fast bacilli growth.

Causes for Rejection

< 6 mL blood in adult tube (strictly enforced); delay in transport; improper labeling; clotted specimen

Turn Around Time

Initial Preliminary report after 2 weeks; Final report at 6 weeks.

Methodology

Culture on selective and non-selective media. AFB isolates are identified at a reference lab.

Patient Prep

Skin prep same as for blood cultures.

Post Processing

Observe for complications of venipuncture.

Reference Range

No acid fast bacilli isolated after 6 weeks.

Limitations and Contraindications

Not all mycobacteria will grow on routine culture.  Notify laboratory if M. bovis, BCG, M. haemophilum, or M. leprae is suspected.

Interpretation

 

Additional Information

Reference laboratory performs susceptibility test of initial M. tuberculosis isolates automatically. For other mycobacterial susceptibility tests, contact the Microbiology Laboratory.
